If a misfire is cylinder-specific, move one component at a time when practical—plug, coil or injector—and see whether the problem follows. If it stays with the cylinder, mechanical condition, wiring or cylinder-specific fuel/air issues move higher on the list.
A swapped ECU needs a trustworthy timing reference. When the engine-management system allows it, verify commanded versus actual ignition timing using the procedure appropriate to that ECU/engine. Trigger offset errors can make a tune look reasonable on a laptop while actual timing is wrong.
Cranking voltage and grounds matter because trigger and coil behavior are most stressed during startup. Record battery voltage while cranking and voltage drop across major power/ground paths before replacing sensors based only on an intermittent no-start symptom.
